The sulfation of chondroitin sulfate in embryonic chick cartilage epiphyses.

چکیده
With the use of purified chondroitinase of Proteus vulgaris, a method has been devised for the determination of relative amounts of chondroitin, chondroitin-4-SO*, and chondroitin6-SO4 in cartilage. This method was applied to the analysis of embryonic chick epiphyseal cartilage. With 35S04 and ‘*C-acetate it was possible to study the relationship of embryo age to rate of formation of chondroitin, chondroitin-4-SO*, and chondroitin-6-SO*. Between 10 and 19 days there is a progressive decrease in synthesis of chondroitin and chondroitin-4-SO4 and a progressive increase in synthesis of chondroitin-6304.

Copolymers of chondroitin 4-sulfate and chondroitin 6-sulfate in chick embryo epiphyses and other cartilage.
Tetrasaccharides were prepared from chondroitin sulfate by means of a limited degradation with chondroitinase ABC. Tetrasaccharides containing one sulfate per disaccharide unit were isolated and were found to be of three types: a tetrasaccharide with two 6-sulfated disaccharide units, a tetrasaccharide with two 4-sulfated disaccharide units, and a tetrasaccharide with one 4-sulfated disaccharid...

Microsomal preparations from chondroitin 6-sulfate-producing chick embryo epiphyseal cartilage, and from chondroitin 4-sulfate-producing mouse mastocytoma cells, were incubated with UDP-[14C]glucuronic acid and UDP-N-acetylgalactosamine to form non-sulfated proteo[14C]chondroitin. Aliquots of the incubations were then incubated with 3'-phosphoadenylylphosphosulfate (PAPS) in the presence or abs...
متن کاملAlterations of chondroitin sulfate synthesized by chick embryo cartilage cultured in the presence of 6-aminonicotinamide.
Treatment of day-4 chick embryos with 6-aminonicotinamide (6-AN) impairs limbs chondrogenesis and produces micromelia. Interference with limb cartilage development may be related to decreased NAD-dependent synthesis of ATP due to the fact that chondrogenesis is dependent upon anaerobic metabolism. To better understand the effect of 6-AN on chondrogenesis, isolated cartilage epiphyses from day-1...

A microsomal preparation from chick embryo cartilage has been shown to catalyze the incorporation of sulfate-36S from 3’-phosphoadenosine 5’-phosphosulfate-3% into microsomal glycosaminoglycan. This enzyme preparation is the same preparation that previously has been shown to catalyze the formation of chondroitin (nonsulfated) from sugar nucleotide precursors. The microsomal sulfate acceptor app...

genesis, mechanism of the actions of the hormone on skeletal growth has been under study for many years. Despite these extensive studies the mechanism remains still obscure. Ellis et al.(1953) showed that the incorporation of radiosulfate into cartilage of immature growing rats was greatly reduced by hypophysectomy and restored by growth hormone administration. Denko and Bergenstal (1955) also ...
